Chemical UVB filter. Often combined with avobenzone to broaden protection. Limited to 10% in the EU due to potential endocrine activity.
Topical application
BLimited evidence. One RCT or several controlled studies with limitations.
FDA approved as a UV filter. SCCS lowered maximum concentration to 7.34% (2021) due to in vitro endocrine activity data. Systemic absorption confirmed in FDA studies – plasma concentrations exceed the threshold requiring additional toxicological data.
Homosalate is a synthetic UVB filter, the salicylate ester of 3,3,5-trimethylcyclohexanol. Covers UVB (absorption peak 306 nm) with no UVA activity. Used as a co-filter to boost SPF in formulations. Where applied. Sunscreens up to 10% in EU (SCCS 2021 lowered the cap from 15% to 10% over endocrine concerns), up to 15% in US, up to 10% in Canada. Often combined with octisalate, avobenzone, octocrylene for broad-spectrum coverage. EU 2024 restrictions. SCCS reviewed homosalate in 2021-2024 and reduced the maximum concentration in leave-on products from 15% to 7.34% (effective 1 January 2025) over suspected endocrine activity. Up to 10% remains permitted on the face; no limit in decorative cosmetics. In the US, FDA's 2019 TEA placed homosalate in category III (additional safety data required) but did not ban it. Safety. Systemic absorption through intact skin was reported in FDA Matta (JAMA 2020). In vitro endocrine tests showed weak activity as estrogen receptor agonist, androgen receptor antagonist, and 17-β-HSD3 inhibitor. In vivo data on clinical significance are limited. Pregnancy and lactation – use with caution. SCCS 2024 did not ban it but recommends limiting exposure. In pregnancy and lactation, mineral filters (zinc oxide, titanium dioxide) or newer organic filters (Tinosorb S, Mexoryl 400) are preferred. Alternatives. Mineral filters, Tinosorb S/M, Mexoryl SX/400, octyl methoxycinnamate.
