Acetyl Decapeptide-3 (trade name Rejuline by Lipotec) is a synthetic ten-amino-acid peptide with an N-acetyl group for added stability. Designed as a signal peptide to stimulate fibroblasts. Claimed mechanism. The manufacturer reports activation of collagen and fibronectin synthesis genes in human fibroblast culture: in vitro at 10 ppm it increased cell proliferation up to 40% and type I collagen production up to 73%. Where applied. Anti-aging serums and creams (1–5% of a 10 ppm concentrate). Common in Asian and Spanish pharmacy cosmetics (Sesderma, Mesoestetic). Evidence base. Weak. No independent human RCTs with clinical endpoints (wrinkles, firmness) exist – only in vitro and manufacturer-sponsored work. Marketing extrapolates lab data to skin. Safety. CIR has not assessed the peptide individually, but the SCCS 2018 cosmetic peptide group is considered conditionally safe. Hypoallergenic, non-irritating at typical concentrations. Low systemic absorption through intact skin. Pregnancy and lactation – use with caution. No direct data; biologically active peptides are best avoided as a theoretical precaution. Safe alternatives for gestational firmness loss: niacinamide, adenosine, hyaluronic acid. Realistic take. A peptide with a plausible mechanism and weak clinical base. An add-on to the core routine (SPF, retinoid or bakuchiol in pregnancy, niacinamide, vitamin C), not a replacement.
