Data from two small sponsor-conducted clinical studies demonstrate wrinkle reduction and improved skin firmness. Independent RCTs are absent. In vitro data confirm collagenogenesis stimulation. Actual peptide penetration through the stratum corneum remains debated.

Palmitoyl Pentapeptide-4 (Matrixyl) is a synthetic lipopeptide: the pentapeptide Lys-Thr-Thr-Lys-Ser with palmitic acid. One of the most studied cosmetic peptides, developed by Sederma in 2000. Mechanism. A signaling peptide: a type I collagen fragment that, on collagen degradation, signals fibroblasts to synthesize new collagen. Additionally suppresses matrix metalloproteinases (MMP-1, MMP-3), which degrade collagen. The palmitoyl moiety enables epidermal penetration. Where applied. Anti-aging serums and creams. Concentration 2–8 ppm (0.0002–0.0008%). Known in The Ordinary Matrixyl 10% + HA (actually 100 ppm), Olay Regenerist, Strivectin. In Spain in Mesoestetic, Sesderma C-Vit Glycolic, Skeyndor. Evidence base. Robinson 2005 RCT (n=93) showed 16% reduction in wrinkle depth over 12 weeks with 3 ppm matrixyl. Less effective than retinoids and L-ascorbic acid, but comparable for comfort and compatibility. Safety. CIR confirmed safety. Hypoallergenic, non-comedogenic, does not photosensitize. Pregnancy and lactation – manufacturers advise caution. No direct pregnancy data – synthetic peptides are not studied in detail. AAD 2023 places them as conditionally acceptable but not first-line. Alternatives – adenosine, bakuchiol, niacinamide with stronger evidence base.
