Trifluoroacetyl Tripeptide-2 is a synthetic peptide with a fluorinated N-terminal group. Introduced by Sederma in the early 2000s as the key active in the Matrixyl 3000 complex (alongside palmitoyl pentapeptide-4, now known as Matrixyl Synthe'6). Mechanism – as claimed. The manufacturer describes inhibition of procollagenase and protection of dermal collagen. The fluorinated group raises peptide stability against enzymatic degradation in skin. In cell models (human fibroblasts) it has been shown to upregulate hyaluronan synthase and fibronectin expression. Where applied. Anti-ageing serums and creams labelled 'Matrixyl 3000 inside', eye-area products. Use levels in finished formulas: 0.01-0.05%. In Spain it appears in Sesderma (Reti-Age, Acglicolic), Mesoestetic (Energy C Eye Contour) and Skeyndor lines. Evidence base. Sederma internal studies in 23 women (8 weeks of 3% Matrixyl 3000) showed 30-45% reduction in wrinkle depth. There are no independent RCTs with isolated trifluoroacetyl tripeptide-2; the effect is attributed to synergy within Matrixyl 3000. Safety. CIR assessed Matrixyl-family peptides as safe. Non-sensitising, non-comedogenic, not photosensitising. Pregnancy and lactation – no data. Topical systemic absorption is minimal, theoretical risk is nil, but in pregnancy ingredients with stronger evidence (niacinamide, hyaluronic acid) are usually preferred. Suited for. Mature skin with loss of density, expression lines, anti-ageing prevention in patients aged 30+ as part of a routine with vitamin C, SPF and retinoids.
