Carnosine (L-carnosine, beta-alanyl-L-histidine) is a dipeptide of beta-alanine and L-histidine. The body holds high concentrations in skeletal muscle and nervous tissue. In skin it acts as a natural buffer and antioxidant. Mechanism. Binds reactive carbonyl glycation products (AGEs) – end-products of protein glycation that accumulate with age and hyperglycemia and stiffen collagen. Neutralizes hydroxyl radicals and singlet oxygen. In vitro slows fibroblast aging and lowers keratinocyte oxidative stress. Where applied. Anti-aging serums and creams (0.1-2%), eye-area products, formulas for photoaged or glycation-marked skin (dull tone, yellowish cast). Often combined with vitamin C, niacinamide, and peptides. In Spain – in Sesderma Reti-Age and Mesoestetic Age Element lines. Evidence base. Most data are in vitro and animal-model (Babizhayev 2010). Clinical RCTs on topical use are limited: pilot studies (n<50) report firmness and texture gains over 8-12 weeks, without confirmation in larger independent work. Oral nutraceutical data are better studied but do not transfer directly to topical use. Safety. CIR has not issued a separate review, but carnosine is endogenous, hypoallergenic, and non-comedogenic. Irritation is very rare. Pregnancy – safe at topical concentrations. Systemic skin absorption is negligible. Lactation data are limited, but as an endogenous compound there is no theoretical risk. What to expect. Mild antioxidant effect, support for a comprehensive anti-aging routine. Does not replace evidence-based retinoids and vitamin C. Suitable as an add-on, not as a monoingredient.
