Madecassoside is a triterpenoid glycoside of Centella asiatica, the most studied of the four key TECA-complex components (madecassoside, madecassic acid, asiaticoside, asiatic acid). Structurally it is madecassic acid with an attached sugar, which increases water solubility. Mechanism. Stimulates fibroblast proliferation and type I and III collagen synthesis via TGF-β signaling (Bonté 1994). Suppresses pro-inflammatory cytokines (IL-1β, TNF-α, IL-6) and NF-κB activation. Antioxidant effect via reactive oxygen species neutralization. Accelerates wound healing and epithelialization. Where applied. Post-procedure serums and creams, atopy and rosacea products, repair creams. Concentration 0.1–1%. In Spain in La Roche-Posay Cicaplast, Bioderma Cicabio, Avène Cicalfate, Endocare Cellage Day Prodermis. Often combined with panthenol, niacinamide, ceramides. Evidence base. Hong 2008 clinical study in 60 photoaged patients showed 18% reduction in wrinkle depth over 12 weeks at 0.2%. Bylka 2014 meta-analysis confirmed efficacy of centella extracts (with madecassoside) for wounds, burns, scars. CIR confirmed safety. Safety. Hypoallergenic at cosmetic concentrations, non-comedogenic, does not photosensitize. Allergic reactions are rare (1–3% in eczema and Asteraceae pollinosis patients). Pregnancy and lactation – use with caution. Topical use at 0.1–1% is rated conditionally acceptable. Direct pregnancy data are limited. Oral centella in supplement doses is contraindicated (possible estrogen-like activity). Alternatives – adenosine, panthenol, niacinamide. Who it suits. Sensitive skin, rosacea, post-acne marks, atopic dermatitis, recovery after laser, chemical peels, microneedling. Often combined with panthenol and ceramides for synergistic barrier repair.
