Palmitoyl Tripeptide-8 (Neutrazen) is a synthetic lipopeptide: the tripeptide His-D-Phe-Arg with palmitic acid. An α-MSH hormone analog with anti-inflammatory focus. Mechanism. Binds to MC1R receptors on keratinocytes and suppresses neurogenic inflammation. Reduces CGRP and substance P production – neuropeptides causing redness and burning in rosacea and sensitive skin. Where applied. Serums and creams for rosacea, sensitive skin, couperose, post-procedure recovery products. Concentration 2–8 ppm. In Skeyndor, Sesderma, Endocare Rosatum. Evidence base. Bohbot 2007 RCT (n=20) showed reduced redness and improved skin comfort in rosacea over 4 weeks at 4 ppm. No systematic review. Evidence base is smaller than for brimonidine and metronidazole, but the peptide is in demand in the cosmetic niche. Safety. CIR confirmed safety. Hypoallergenic, non-comedogenic, does not photosensitize. Pregnancy and lactation – topically safe per manufacturers. No direct pregnancy data, but local use at cosmetic concentrations is not systemically significant.
